finding directions from latitude and logitude

Hello. I would like to find directions using lat and long. The numbers are Lat 40.543330 and lon 78593889. Can i enter the numbers in exactly the way they are shown or will i have to know north south east and west.

If the location is Jefferson St in Pittsburgh, then put the data in as N40.543330 and then W78.593889.

i know this is dumb but is the first set of numbers always north and the second set always west?
